There is only one pair of direct high speed trains between Hong Kong and Hefei:  G385 & G386.  It takes about 6-6.5 hours to finish the distance of around 1,267 kilometers (787 miles). The ticket price of a second seat is CNY 858 (US$ 126). 

Alternatively, you can transfer at Shenzhen, Wuhan or Guangzhou for more flexible travel plan. The overall high speed train travel time by this way is about 6.5 - 11 hours and the ticket price of a second class seat is about CNY 701 - 904 (US$ 99 - 128).

 At present, there is only one pair of direct flights between Hong Kong and Hefei on every Monday and Friday, and it takes about 2.5 hours and generally costs more than CNY 800 / US$ 113 for a single journey. So at most times, passengers need to make a transfer in Xiamen, Guilin, Shanghai or other cities, which means flights do not have many advantages over bullet trains in terms of convenience.
